چکیده

Background Monoclonal antibody (mAb) therapy-has changed the natural history of patients with B cell lymphomas, breast cancer, and head and neck cancers. However, response rates are suboptimal, highlighting the need to enhance mAb activity. Combining a tumor targeting mAb with a second antibody that activates natural killer (NK) cells may improve the therapeutic effects of mAbs. Engagement of the Fc receptor (FcR) on NK cells by mAbs coating tumor cells has been previously shown to enhance their expression of CD137 [1-3]. We conducted preliminary analyses as a first step toward developing a nomogram to predict CD137 up-regulation following therapy with mAb.

Characterizing CD137 upregulation on NK cells in patients receiving monoclonal antibody therapy.

Background In the era of personalized cancer medicine, identifying techniques for effectively matching patients to efficacious treatments is a critical step in the treatment process. The advent of anti-cancer immunotherapies necessitates novel approaches to biomarker identification beyond traditional genomic profiling. One promising approach is incorporation of nomograms into treatment decision...

CD137 stimulation enhances the antilymphoma activity of anti-CD20 antibodies.

Antibody-dependent cell-mediated cytotoxicity (ADCC), which is largely mediated by natural killer (NK) cells, is thought to play an important role in the efficacy of rituximab, an anti-CD20 monoclonal antibody (mAb) used to treat patients with B-cell lymphomas. CD137 is a costimulatory molecule expressed on a variety of immune cells after activation, including NK cells. In the present study, we...

Anti-CD137 monoclonal antibody administration augments the antitumor efficacy of dendritic cell-based vaccines.

In weakly and poorly immunogenic tumor models, we examined the effects of stimulating CD137 (4-1BB) in vivo by administering anti-CD137 monoclonal antibody after tumor lysate-pulsed dendritic cell (TP-DC) vaccination. TP-DC subcutaneous vaccination induced a transient up-regulation of CD137 on T cells and natural killer (NK) cells within vaccine-primed lymph nodes (VPLNs). In established pulmon...

Targeting CD137 enhances the efficacy of cetuximab.

Treatment with cetuximab, an EGFR-targeting IgG1 mAb, results in beneficial, yet limited, clinical improvement for patients with head and neck (HN) cancer as well as colorectal cancer (CRC) patients with WT KRAS tumors. Antibody-dependent cell-mediated cytotoxicity (ADCC) by NK cells contributes to the efficacy of cetuximab. The costimulatory molecule CD137 (4-1BB) is expressed following NK and...

Boosting antibody-dependant cellular cytotoxicity against tumor cells with a CD137 stimulatory antibody

Monoclonal antibodies (mAb) induce tumor regression through antibody-dependant cellular cytotoxicity (ADCC). We recently showed that an agonistic anti-CD137 mAb stimulates natural killer (NK) cells which have been activated by a tumor-specific mAb, resulting in increased ADCC against cancer cells.
